To verify the expression of conditional knockin (cKI) mice under normal conditions, the Flox knock-in mice needs to mate with a Cre line to remove the Stop-signal, generating mice that should express the knock-in across the tissues (specified by Cre). If the resulting expression is not as expected, the model will need to be reconstructed - which takes significant time, effort, and resources. The TurboKnockout® gene targeting technique from Cyagen can provide a rapid warning mechanism for early expression verification. When constructing the vector, tag proteins such as Flag or eGFP/mCherry are inserted into the over-expressed genes. After obtaining the ES clones with correct targeting, blastocyst injection will be performed, and ES cells will be subjected to Western Blot to verify the expression of the tag protein. Moreover, compared with CRISPR/Cas9-based techniques, TurboKnockout® is free of patent disputes - making it the technique of choice for new drug development projects.
