1. Sample preparation
We first perform the required breeding needed to obtain transgenic/mutant animals of the desired genotypes. Animals are bred within our SPF facility and sacrificed at specified time points. The tissues/organs are then processed, paraffin embedded, sectioned and stained; For each genotype, a set of tissues will be harvested. Additional samples can be prepared in paraffin blocks upon request.
2. Embeding and sectioning
Embedding and sectioning services are based on standard paraffin sectioning protocosl. Tissues of interest are first fixed and embedded in paraffin blocks; Paraffin blocks can be used for a variety of downstream analyses, including immunohistochemistry (IHC), in-situ hybridization (ISH), RNA-Seq, PCR, etc. Paraffin embedded tissues can also be preserved in blocks for years with good morphology. Paraffin blocks can be further sliced into thinner sections. We can provide paraffin blocks and the unstained sections. By default, a total of three slides are generated from one organ/tissue sample in this service.
For special research purposes like cell lineage tracing that requires serial/step sections, sections collected from different tissue intervals are placed on the same slide to present a full picture of the tissue.
3. Hematoxylin and Eosin (H&E) staining
As the most commonly used histologic staining approach, H&E staining provides a detailed view of the tissue for routine examination. While hematoxylin stains nuclei into blue/purple, eosin counterstains cytoplasm, muscle and connective tissue into pink/red.
4. Imaging
We currently provide imaging services for the gross morphology and H&E staining. Images will be labeled and stored on our FTP site for download; Data can be accessed anytime and shared with collaborators instantly. We provide three months of FTP data storage for free, and can provide longer storage times upon request.
