Case Study

Cyagen has generated a number of conditional knockout mouse models with the using of CRISPR technology and the Cre-Loxp system for Cre conditional knockout (cKO). In this case, gene Slc30a7 conditional knockout mouse line was created in the C57BL/6N background and subsequently crossed with the proper Cre animals. The strategy of gene Slc30a7 cKO mice was to select exon 4 and exon 5 as conditional knockout regions. CRISPR technology was used for insertion of two LoxP sites flanking the cKO region for conditional removal using Cre conditional knockout technology.

Where do your mice come from?
Our main source of mice is Charles River Laboratory. Currently, only mice from Charles River laboratory, Jackson Laboratory and Taconic are allowed into our facility.
How long does it take from DNA microinjection to obtaining transgenic animals?
Typically, it takes about 8 weeks from receipt of DNA to shipment of first-generation mice: 1-2 weeks to complete the microinjection process, around 3 weeks for pups to be born, and 3-4 weeks before mice are weaned and shipped from our facility. The quality of the DNA preparation and the accuracy of the concentration detection can significantly affect the project turnaround, so we will request that DNA that does not meet our criteria be replaced. For those services requiring only transgenic embryos, the time can be as short as 4-5 weeks. However, if the number of injections needs to be increased, the project turnaround may be extended as appropriate.
What is your success rate in generating first-generation transgenic founders? Is there a guarantee?
The success rate of generating first-generation transgenic mice with plasmid DNA is approximately 3-4% (i.e., approximately 5-8 PCR-positive first-generation mice can be obtained for each injection of 200 zygotes). We promise to provide at least two PCR-positive first-generation transgenic mice. We guarantee at least two first-generation mice that carry some or all of the transgene, but we do not guarantee expression of the transgene or germline inheritance. If the transgene is embryonic lethal or the DNA provided by the customer does not meet the requirements, the customer needs to understand that they will bear the full cost of the microinjection. Due to the lower success rate of BAC, there is no guarantee on first-generation transgenic mice, however, customers can choose to add an additional round of injections to improve their chances of success with our discounted option.
