The experimental principal is to allow cells to grow to a single layer of fusion, then artificially create a blank area (also called a "wound") on these cells. The cells on the edge of the space will gradually enter the blank area to close the wound.

 

Compared to other methods, the in vitro wound/scratch assay simulates the process of in vivo cell migration to a degree, and is the simplest among in vitro testing methods for studying cell migration.
