In vivo tumor models, such as cell line-derived tumor xenograft (CDX) models, play an important role in basic research of tumorigenesis and metastasis, as well as drug development and treatment. Therefore, establishing a precise in vivo animal model is vital for accurate tumor studies and in advancing oncology research.

Tumor cells are widely used in establishing tumor models due to its ease of access and low cost. In addition, there are many publications and data on genomics, cell function, and pharmacodynamic responses available for reference.

What is a Cell Line-Derived Tumor Xenograft (CDX) Model?

A cell line-derived tumor xenograft (CDX) model is constructed by transplanting human tumor cells into immunodeficient mice. Human tumor cells are cultured in vitro before they are implanted into immunodeficient mice. CDX mouse models are typically used to observe the growth of tumor, tumor cell proliferation, and in vivo drug screening.

Types of CDX Mouse Models

(1) Subcutaneous CDX Model:

Tumor cells are implanted into the subcutaneous area of the armpit or back of the forelimb of nude mice to observe the tumorigenesis results. It is the most commonly used model in tumorigenesis research.

The Advantages of Subcutaneous CDX Model:

  • The process is simple to operate, which can directly observe the growth of tumor and detect important data such as animal weight, tumor growth curve and tumor weight.
  • It is widely used in biomedical research and drug development.

(2) Orthotopic CDX Model:

Tumor cell lines are implanted into in situ organs or tissues of mice for tumor bearing.

The Advantages of Orthotopic CDX Model:

  • Better simulate the microenvironment of tumor cell growth in vivo.
  • Simulate the process of tumor growth and even metastasis.
